IDE inhibitors for the treatment of diabetes
Technology Description Current therapeutic strategies for managing diabetes include increasing insulin levels by direct injection, stimulating insulin production, and improving insulin effectiveness. Modulating insulin degrading enzyme (IDE) activity represents another potential means by which insulin levels may be increased. IDE is the principle enzyme responsible for the degradation of insulin and is expressed ubiquitously. Although IDE has been studied for more than 60 years, the discovery of IDE inhibitors has been hampered by the unique conformation of the enzyme active site. Mayo Clinic researchers have developed unique and sensitive assay methodologies to facilitate compound library screening for IDE inhibitors. Multiple classes of small molecule compounds have been identified that inhibit IDE activity. Studies using some of the IDE inhibitors demonstrated favorable regulation of insulin catabolism and signaling, consistent with therapeutic utility for diabetes.
Application IDE inhibitors could potentially be used to increase circulating insulin levels and potentiate insulin signaling for the treatment of diabetes.
Stage of Development Multiple classes of small molecule inhibitors have been identified and additional characterization and structure-activity relationship (SAR) studies are ongoing.
